Indeed, the first pharmacopoeia of the United States, now in its 34th revision, provided recipes for crude mixtures drawn from nature. It was only later that the promise of medicines as chemicals, drawn from the precepts of Paracelsus, came to dominate the therapeutic armamentarium for medicines of the 20th century.

The Polo like kinases play key roles in mitosis. While the upregulation of PLK1 in cancer is well documented and PLK3 has been demonstrated to be a tumor suppressor, little is known about the oncogenic significance of PLK2. PLK2 kinase activity is essential for centriolar duplication and is also believed to play a regulatory role in the survival pathway by physically stabilizing the TSC1/2 complex in tumor cells in hypoxic conditions.

I will assess this likely evolution and fragmentation of the biosimilar market in my presentation. I believe that this richness of options would lead it to be a much larger market than the biosimilars itself. There will be regulatory challenges and they would be overcome on case-by-case basis depending on the quality of the science behind a particular approach.

Cancer stem cells are a very small subset of cancer cells and possess characteristics very similar to normal stem cells, in particular, the capacity for self-renewal, multipotency and relative quiescence. These chemo- and radiation resistant cells are responsible for maintaining tumor volume leading to therapy failure and recurrence. This necessitates alternate treatments to existing cancers and many novel treatment modalities are being investigated to specifically target this small group of cells.
